NAD⁺ / NMN (LE Optimized NAD+ NMN + Resveratrol Elite)

NMN (NAD⁺ precursor) + Trans-Resveratrol in a single capsule · Mitochondria and sirtuins · Chronic (8–12 wk)

Evidence C · Moderate Fat-soluble (Resveratrol) Chronic (8–12 wk)

NMN (β-nicotinamide mononucleotide) is the direct precursor of NAD⁺ — the central coenzyme of mitochondrial respiration, DNA repair (PARPs) and the sirtuin metabolic sensors (SIRT1/SIRT3). NAD⁺ declines with age and with high training volume, and replenishing it is the most promising hypothesis for sustaining mitochondrial function. This product combines NMN with Trans-Resveratrol (a direct SIRT1 activator) — the most-investigated pair in the literature: NMN provides the substrate, resveratrol activates the enzyme that uses it.

Why it matters for ultramarathon runners

How it works (mechanism)

NMN enters the cell via the Slc12a8 transporter or is converted to NR (nicotinamide riboside) extracellularly and absorbed. Inside the cell, NMN is converted to NAD⁺ by the enzyme NMNAT. NAD⁺ feeds the sirtuins (SIRT1 activated by resveratrol = perfect combo), PARPs (DNA repair) and the mitochondrial respiratory chain. SIRT1 activated → deacetylates PGC-1α → mitochondrial biogenesis + better β-oxidation.

Product: LE Optimized NAD⁺ NMN + Resveratrol Elite

Optimized NAD+ NMN + Resveratrol Elite (Life Extension) combines both actives in a single capsule:

⚠️ Overlap with your stack: this combo already contains Trans-Resveratrol. If you use this product, the standalone LE Optimized Resveratrol Elite line in the summary becomes redundant — consider dropping the isolated version so you don't double up on resveratrol and cost. Same logic as the LE Mitochondrial Energy Optimizer w/ PQQ, which covers PQQ + R-ALA in one capsule.
