NMN (β-nicotinamide mononucleotide) is the direct precursor of NAD⁺ — the central coenzyme of mitochondrial respiration, DNA repair (PARPs) and the sirtuin metabolic sensors (SIRT1/SIRT3). NAD⁺ declines with age and with high training volume, and replenishing it is the most promising hypothesis for sustaining mitochondrial function. Why it matters for ultramarathon runners
- NAD⁺ fuels the mitochondria: electron carrier in Complexes I and III of the respiratory chain. Without enough NAD⁺, ATP falls and recovery stalls.
- High training consumes NAD⁺ faster: volume and intensity raise NAD⁺ turnover via PARPs and oxidation. Replenishment via precursor (NMN) is the most robust strategy demonstrated in humans.

How it works (mechanism)
NMN enters the cell via the Slc12a8 transporter or is converted to NR (nicotinamide riboside) extracellularly and absorbed. Inside the cell, NMN is converted to NAD⁺ by the enzyme NMNAT. NAD⁺ feeds the sirtuins (SIRT1 activated by resveratrol = perfect combo), PARPs (DNA repair) and the mitochondrial respiratory chain. SIRT1 activated → deacetylates PGC-1α → mitochondrial biogenesis + better β-oxidation.
NMN vs. NR vs. Niacinamide
- NMN: closest precursor to NAD⁺ (1 enzymatic step). Human studies growing since 2020. More expensive.

⚠️ When NOT to use / cautions
- Cancer history/risk: NAD⁺ is "food" for rapidly replicating cells too. In people with active cancer or recent history, discuss with an oncologist first — the literature is still controversial.
